Why Can I Not Sleep on GLP-1 Medications?
Sleep disruption is commonly reported on GLP-1 medications, and the reasons are not fully established. Two contributors are often discussed: reduced tryptophan intake from eating substantially less, and the physiological demands of rapid weight change. Why can I not sleep on GLP-1 medications?
Sleep disruption on GLP-1 medications is commonly reported and rarely discussed in advance. Patients describe a recognisable pattern: exhausted during the day, unable to fall asleep at night, waking frequently, never feeling rested. Why it happens is not fully established, but there are plausible physiological explanations worth understanding.
A proposed mechanism: tryptophan
The body produces serotonin from tryptophan, an amino acid obtained through dietary protein. When food intake drops significantly, and a 2025 joint advisory from four professional societies (Mozaffarian et al., Obesity) reported reductions in caloric intake of 16 to 39 percent across the GLP-1 drug class, tryptophan intake may drop with it. Because serotonin is involved in sleep onset, this is a proposed explanation for why falling asleep becomes harder. It is plausible and mechanistically grounded, but it has not been demonstrated in GLP-1 users, and we say so. A similar mechanism is discussed for mood changes on GLP-1 medications.
A second proposed contributor: the demands of rapid change
Rapid weight loss places real demands on the body, and some researchers discuss stress physiology, including cortisol, as a contributor to feeling alert at night when you should be winding down. Changed eating patterns and smaller evening meals are also discussed. None of these has been isolated as the cause in GLP-1 weight loss specifically. Most likely several factors overlap.
The distinction from fatigue
GLP-1 sleep disruption and GLP-1 fatigue are related but different problems that often co-occur. Fatigue is primarily discussed in terms of nutritional intake and cellular energy. Sleep disruption is about what happens at night. Attending to one does not automatically resolve the other.
What the evidence supports
Melatonin. Involved in regulating the sleep-wake cycle, and the ingredient in TAKE Rest present at a meaningful dose. Rest provides 5mg per capsule, with a label serving of two capsules (10mg). That is a high dose by the standards of most melatonin research, where 0.5mg to 5mg is typical, so start with one capsule.
L-Tryptophan. The first named ingredient in the 905mg proprietary blend, and the precursor to serotonin. Its inclusion follows the reasoning above, that dietary tryptophan may be reduced during GLP-1 treatment. The mechanism is plausible but has not been tested in GLP-1 users. The rest of the blend. Rest also contains lycium, chamomile, lemon balm, passion flower, L-taurine, hops, St. John's Wort, GABA, Chinese skullcap, L-theanine, ashwagandha, inositol and 5-HTP. These sit within a single proprietary blend and are listed in descending order, so the ingredients near the end of that list, ashwagandha and 5-HTP among them, are present in small amounts. We do not headline them and you should not expect them to behave like the doses used in trials of those ingredients on their own.
